Infectious Diseases has a requirement to procure: a peptide library
consisting of 2,054 19-mers. This is intended to be procured on a sole
source basis. Mimotopes is the only distributor of Cleavable Peptide
Libraries consisting of the following features: (1) The individual
peptides must be synthesized using multiple parallel synthesis on pins
in 96-sample blocks using multipin solid phase peptide synthesis
technology to minimize costs and turn around time. (2) The resultant
peptides must be biotinylated at the aminoterminus and must be
synthesized with a 4-residue (S-G-S-G) spacer arm. The synthesis
technology must allow the peptides to be cleaved after synthesis on
pins, synthesized in 1-5 mmol quantities, and supplied in the final
form as lyophilized powder. In addition, Mass Spectral Analysis must be
performed on selected peptides as part of the quality control process.
(3) Mimotopes is able to synthesize at least 1,000 peptides per week
making it possibleto supply the peptide libraries within 4-5 weeks of
the placement of the order. 4) Cleavable peptide libraries can be used
to detect antibody/peptide interactions with the goal of mapping all
of the epitopes (regions of a protein recognized by antibody) in a
particular protein. They are an ideal way of searching through protein
antigen sequences for the location of B-cell or helper or cytotoxic
T-cell epitopes. Focusing on the studying of genes contributing to the
virulence of Group A Streptococcus (GAS). The cleavable peptide
libraries will identify the regions of presumptive virulence proteins
that are recognized by human antibody. The results will lead to
insights about the functional sites within these proteins, their roles
during GAS infection, and may ultimately lead to successful vaccine
strategies. Closing date for the submission of written responses is
