Government intends to negotiate a modification to the statement of work
for the contract with Howard University College of Medicine. The
contractor shall be required to (1) enroll five additional family
members of the affected sib-pairs for a total of 2000 persons. Each
West African site shall be required to enroll 400 persons. Offspring
participants shall be above the age of 14; (2) perform high resolution
mapping and candidate gene analysis. A core set of candidate genes for
type 2 diabetes shall be analyzed on all samples. Allele frequencies
and haplotype analyses shall be performed on the candidate genes.
Single Nucleotide Polymorphisms (SNP) technologies and linkage
disequilibrium mapping shall be used by the NHGC Molecular genetics
Laboratory to generate databases for functional mutation analyses in
candidate genes; (3) perform statistical genetic analyses and
bioinformatics. Using the broadly applicable distributions, and the
classification and regression trees (CART) method for screening large
numbers of markers and other risk factors, modeling and distribution
and familial correlations in age-of-onset; Segregation and linkage
analyses; Implementation in the software package, G.E.Ms, which is
designed to be user-friendly and portable across common computing
platforms. The changes to the statement of work will not require an
extension of the expiration date of the contract. The current employees
will increase their level of effort. See Numbered Notes 22 and 26.
