Standards and Technology (NIST) intends to procure, on a sole source
basis, the services of Susan Tai, of 822 Bald Cypress Drive, Rockville,
MD 20850 under the authority of FAR 13.106-1(b) Only one source
reasonably available. This procurement is for complete development and
validation of LC/MS method for total serum thyroxine and serum
cortisol. The contractor will complete method development for the
determination of total serum thyroxine and cortisol using LC/MS and
will rigorously evaluate the accuracy and precision of the method to
assure that it meets the high standards of NIST. The method will be
applied to selected materials for measurement. The determination of
free thyroxine, representing about 0.1% of the total thyroxine must be
investigated to determine if a method can be established to isolate
free thyroxine from protein-bound thyroxine. The method will be applied
to the determination of cortisol in selected materials. The method
development will include extractionof thyroxine and cortisol from serum
in high yield and without decomposition, further processing of the
extract to remove potential interferences, liquid chromatographic
separation, and electrospray ionization mass spectrometry for
measurement. The contractor must have demonstrated skill in developing
high accuracy methods for trace level analytes in biological matrices
and must have expertise in the chemistry of thyroxine and cortisol, in
organic isotope dilution, in modern liquid chromatography, and in
electrospray ionization mass spectrometry. The methods must be tested
by repeated measurements of thyroxine in pools of human serum with
thyroxine levels that are representative of the range encountered in
normal and thyroid-impaired human populations, and of cortisol in pools
of human serum with cortisol levels that are representative of the
range encountered in normal human populations as well as those with
elevated or depressed endocrine function. Alternate measurement
strategies must be devised to test for bias in the measurements.
Comparison with field method results will be included to assure that
there is commutability between the methods. The methods must be applied
to the determination of thyroxine and cortisol in SRM materials and any
other serum materials that the COTR deems necessary. This is a follow
on project where Susan Tai worked under a CRADA that has expired, with
the College of American Pathologists performing the same tasks. Susan
Tai has knowledge and experience in all of the areas mentioned. She
understands the stringent accuracy and precision requirements that are
expected of NIST methods and measurements. Award to another source
would result in substantial duplication of cost that the Government
would not recover through competition and would create unacceptable
delays in fulfilling NIST's mission requirements. It is anticipated
that a Firm Fixed Price Purchase Order will be issued by September 11,
2000. The Period of Performance is for one year from the date of
award. Interested persons may identify their interest and capability to
respond to the requirement. This notice of intent is not a request for
competition. However, the Government will consider all responses
received within 15 days after the date of publication of this synopsis.
Written responses to this synopsis shall contain sufficient
documentation to establish a bonafide capability to fulfill the
