26505-2888 Q -- GENERATION OF A KNOCKOUT MOUSE MODEL SOL RQ2000-034 DUE 082500
POC Brenda Goodwin, 304/285-5882; Contracting Officer, Rex S. Wolfe,
Jr., 304/285-5883 NOTICE OF INTENT TO SOLE SOURCE The National
Institute for Occupational Safety and Health (NIOSH), Toxicology and
Molecular Biology Branch, HELD in Morgantown, WV is currently studying
the function of a novel gene (T1G1) that is inducible by dioxin and
may contribute to the etiology of dioxin toxicity. Current analysis of
the gene suggests that T1G1 is regulated during tumor formation in
lymphoid tissues and memory processes in the brain. Generating a knock
out mouse model in which the gene of T1G1 is altered or deleted would
permit genetic study of the physiological function and the role in
dioxin toxicity of T1G1. This work requires a company specialized in
gene targeting with state of the art equipment and technology. The only
resource known to have the capabilities to generate a knockout mouse is
inGenious Targeting Laboratory, Inc., Stony Brook, NY. This corporation
will perform cell work, electroporation, and screening of clones until
identification and confirmation of positive ES clones is accomplished.
This corporation guarantees a successful germline transmission once a
positive ES clone is identified and provides F1 heterozygous mice
instead of the chimera founder mice. Once the gene targeting services
for this project are performed and the F1 heterozygot is obtained, the
gene targeting laboratory has no ownership to the animals and no
royalties are ever collected. If germline transmission is not achieved,
the company does not require payment for performance of unsuccessful
gene targeting services. The facilities house animals in maximum
isolation and every item that enters the isolation rooms is autoclaved.
Mice are maintained under specific pathogen-free conditions and
serology tests are performed every 6 weeks where health reports are
provided. There is no other known source available, however, if there
are vendors having the capabilities necessary to meet the stated
requirements, they are invited to submit appropriate documentation,
literature, brochures and references to Brenda L. Goodwin, Reference:
RQ2000-034, DHHS, PHS, CDC, NIOSH, ALOSH, 1095 Willowdale Road,
Morgantown, WV 26505. The Government intends to negotiate on a
sole-source basis with inGenious Targeting Laboratory, Inc., Stony
Brook, NY. Interested organizations must provide a description in
sufficient detail to show that the services being offered demonstrate
adequate qualifications/capabilities and a successful record of past
performance. To be considered, responses shall be submitted within 14
days after publication of this notice. Information received will
generally be used for the purpose of determining whether to conduct a
competitive procurement. This notice is issued for informational
