Remsburg Jr., Purchasing Agent (301) 402-2282/John Foley, Contracting Officer (301) 402-2284 17) The National Institute of Allergy and Infectious Diseases, NIH has a requirement for and is seeking business concerns for screening several cDNAs in a yeast two-hybrid system for interaction against a large library of human cDNAs. Each cDNA will be supplied in a plasmid vector. Work to be performed by the contractor will include the following: 1.) Prepare each provided cDNA to be used as "bait". If possible, the cDNA would be engineered as both N-terminal and C-terminal fusions. 2.) Optimize the selection to eliminate false positives or employ a multi reporter system to minimize false positives. 3.) Screen against a large library of human cDNAs (> 106 independent clones,) Preferably from a HeLa cell or respiratory tract-derived cell. 4.) Identify positives (candidates.) 5.) Phenotypic screening to assure candidates are positive, and validation of clones. 6.) Candidates would be provided as plasmids cloned in E. coli.
