Description

The John D. Dingell VA Medical Center is seeking sources to provide crystallography research trials to be utilized in a research project to define the factors involved in the deactivation of aminoglycoside antibiotics by phosphotransferase enzymes expressed by drug-resistant bacteria, by X-ray crystallographic analysis of several enzymes and enzyme-drug complexes, and to compare and contrast the determinants of drug binding and deactivation in the different classes of aminoglycoside-modifying enzymes, in order to provide a molecular basis for the design of inhibitors or novel animoglycoside compounds. Equipment necessary to conduct these trials must include: Mar345 Image Plate Detector, which uses a Rigaku rotating anode X-ray source, and diffraction data from native crystals will be collected, along with any heavy metal derivative data that might be required to solve the structures. Also needed, direct access to a synchrotron radiation source that used high-intensity X-rays to obtain high-resolution diffraction data.
