- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued. This procurement is being issued as a Request For Quote (RFQ). Submit offers on RFQ1002. The request for quotation (RFQ) and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 97-20. This acquisition will be processed under Simplified Acquisition Procedures (SAP) and is not a small business set-aside. The National Institute of Allergy and Infectious Diseases intends to procure Gene Knockout Services. The purpose is to allow efficient and timely generation of transgenic animals that are deficient in the gene RGS4 for analysis by the Molecular Signal Transduction Section. A mouse will be created that lacks the gene encoding RGS4 (an RGS4 knock-out mouse) and replace a portion of the RGS4 gene with a lac Z reporter construct that will confer information regarding expression time and location. The Government needs to know by phase(s) the entire mouse knockout project, from the cloning of the mouse gene to germline transmission. The company will create a plasmid vector that will produce a targeted deletion of RGS4 in mouse embryonic stem cells while simultaneously allowing selection of targeted cells and substitution on the LacZ gene. The contractor shall submit the vector strategy for the client's approval prior to subsequent steps. The contractor will introduce the vector into embryonic stem cells by electroporation, and the contractor is responsible for determining the genotype of transfected cells. The contractor will then inject 2-4 targeted ES clones into blastocysts for the generation of chimeric mice. The contractor will then cross the offspring and determine if germline transmission of the mutant allele has occurred. Once mice are produced that are heterozygous for the RGS4 mutation, the contractor shall give the mice to the laboratory of allergic diseases without retention of any rights or patents on the mice. The contractor is expected to provide regular monthly progress reports on the development of the mice and report any technical problems or irregularities promptly. The National Institutes of Health will provide to the contractor all information pertaining to the isolated and sequenced genomic DNA encoding RGS4. Offerors must include with their offer a completed copy of provisions at FAR 52.212-3 Offerors Representations and Certifications Commercial Items. FAR 52.227-17 Rights in Data Special Works (Jun 1987) is applicable. The Government will award a fixed price purchase order to the responsible contractor, price and the following factors will be considered: (a) From inception to final delivery shall be nine to twelve months or sooner. (b) Documented methods proven in generating mice. (c) House and care for all animals in compliance with the U.S. animal welfare regulations; clean facility. (d) The ability to include the LacZ reporter gene is essential for this construct. (e) Government shall retain rights to use and freely distribute the gene knock-out mouse for research and development purposes. (f) Guarantee of germline transmission.
