Description
The USDA, Agricultural Research Service, US Meat Animal Research Center (MARC), Clay Center, NE has a requirement for three (3) normalized cDNA libraries (MARC 6BOV, MARC 7BOV, and MARC 3PIG). Specifications for preparation of MARC 6BOV, MARC 7BOV, and MARC 3PIG libraries: Materials supplied by MARC will be a set of 5 tissue samples for each library. These should be homogenized prior to individual mRNA extraction, as some represent tissue pools from multiple individual animals in different physiological states. After mRNA purification, like amounts of mRNA from each tissue shall be pooled prior to cDNA synthesis. First strand cDNA synthesis should be performed using anchored poly(T) primers to provide uniform length of T sequence when sequencing from the 3' end of the clones, or other steps to reduce or remove long stretches of T that interfere with efficient 3' end sequencing shall be employed. The cDNA shall be directionally cloned into a vector for which proper licensing is available to the supplier, and which contains the following features: (1) bacteriophage polymerase promoter sites for production of sense and antisense RNA probes. (2) mammalian expression promoters, preferably inducible. (3) drug resistance selectable markers that allow selection in both bacterial and eukaryotic cell line systems. (4) high copy number origin of replication. Primary libraries must have average insert sizes no less than 1.5 kb, and contain > 10 to (6th degree) clones. Any amplification steps required must use an approach such as semi-solid agarose to minimize clone bias during amplification. This acquisition is being conducted on a "best value" procurement using commercial items procedures. Evaluator factors are price, quality, past experience, proof of meeting the characteristics in the statement of work. Proposals shall provide information regarding the vector to be used; quality control measures; request to retain portions for commercial use and the acknowledgment of the above release process; and the price quotation.
